This one-day workshop will describe some of the fundamental theory underpinning polygenic risk score (PRS) analyses, provide hands-on practical sessions illustrating both basic and more complex PRS analyses, and illustrate the application of PRS using examples from published and ongoing research.
We will use our PRSice software in computer practicals but will also describe how the same kind of analyses can be performed with alternatives (eg. LDpred). Topics covered will include: QC, pruning/clumping, the sample overlap problem, estimating pseudo-R2, ascertainment, multi-ethnic PRS, over-fitting, shrinkage methods, PRS for G*G/G*E and the potential for PRS to contribute to clinical prediction and Personalised Medicine.
