Analytical Bites: Tandem Quadrupole MS and MS Imaging For Investigation of PharmacoMetaboDynamics & Off Target Pharmacology

Join us for the first in our new Analytical Connections series: Drug Development & Target Interactions, exploring how advanced LC-MS/MS and MS imaging are transforming early-stage drug discovery.

Understanding the fate and effect of a candidate pharmaceutical on a biological system is a key part of the drug discovery and development process. DMPK and biomarker studies provide information on ADME, generation of potentially toxic metabolites, and dysregulation of metabolic processes, forming an essential part of the drug development package. Despite these studies, candidate drugs are often eliminated in preclinical and early clinical phases due to toxicity or undesirable DMPK properties. The combination of microsampling and the exquisite sensitivity of modern LC/MS/MS and MS imaging has the potential to rapidly produce significantly more information from early discovery samples, improving the candidate selection process. This presentation will illustrate the use of discovery and targeted LC-MS/MS as well as MS imaging to investigate the relationship between drug-metabolite exposure and changes in endogenous metabolite-lipid levels in two in vivo studies, a TKI inhibitor and an antihistamine hepatotoxin.

Robert Plumb

 Dr. Robert Plumb, Scientific Advisor, Waters Corporation

Dr Plumb is currently a scientific advisor in the Research Development Analytical Testing organization at Waters Corporation. He has several years of experience in the pharmaceutical industry beginning in 1987 at Glaxo R&D DMPK Department focussing on regulatory bioanalysis and metabolite identification. In 2001 he moved to Waters Corporation in Milford, MA, USA where he has worked in R&D, pharmaceutical business development, metabolic phenotyping and discovery omics. Research interest includes application of fast chromatography and advanced mass spectrometry in bioanalysis and omics studies. Dr Plumb has published over 150 papers on LC/MS(MS) and NMR for bioanalysis, metabolomics and metabolite identification.
